Spinach chloroplast RNAs have been translated in the cell-free systems prepared from E. coli, rabbit reticulocyte and wheat germ. Production of the large subunit of ribulose-1,5-bisphosphate (RuBP) carboxylase has been demonstrated by mono- and two-dimensional gel electrophoresis and partial proteolytic digestion in the E. coli and rabbit reticulocyte systems only. However, differences were consistently observed in the overall two-dimensional patterns of the translation products obtained in the above mentioned systems as well as in comparison with the pattern obtained in organello. Similarly, considerable differences were evident when the total RNA population (cytoplasmic plus organellar) was translated in the same systems.
