A purine nucleoside phosphorylase from Aeromonas hydrophila was cloned, expressed in Escherichia coli and purified. The enzyme showed a good stability and a broad substrate specificity towards modified purine ribonucleosides. The enzyme was used for the synthesis of purine nucleosides structurally related to compounds of biological interest via a “one pot, one enzyme” transglycosylation reaction using 7-methyl guanosine iodide as the sugar donor and 6-substituted purines as acceptor.
